硝酸甘油对持续心肌缺血模型动物心肌细胞凋亡及凋亡相关基因的影响

摘  要:目的:研究硝酸甘油对持续心肌缺血心肌细胞凋亡以及对凋亡相关基因的影响。方法:采用大鼠动物模型,用原位标记与半定量分析检测心肌细胞凋亡程度;以S-P免疫组化分别检测Fas基因蛋白。结果:持续心肌缺血组与假手术组相比,心肌细胞凋亡程度明显增加,Fas-L蛋白、Bcl-2蛋白的表达都有明显增高,差异有极显著性(P<0.01);硝酸甘油组与持续心肌缺血组比较,心肌细胞凋亡程度明显降低,差异有极显著性(P<0.01);Bcl-2蛋白表达增高,差异有显著性(P<0.05)、Fas-L蛋白表达降低,差异有显著性(P<0.05)。结论:硝酸甘油能够减轻持续心肌缺血过程中心肌细胞凋亡程度。OBJECTIVE To study the effect of nitroglycerin on apoptosis of myocardial cells and gene expression due to apoptosis during continuous ischemia. METHODS The degrees of myocardial apoptosis were identified by in situ end labeling and semi-quantitative analysis, Fas-gene protein was detected by S-P immunochemistry. RESULTS Comparing continuous ische mic group with sham operation group, the degrees of myocardial apoptosis were increased obviously in continuous ischemic group, the Fas-L proteins and Bcl 2 proteins expression were significantly raised, there was significantly difference(P〈0. 01 ) ; Comparing nitroglycerin group with continuous ischemic group, the degree of myocardial cells apoptosis was obviously reduced in nitroglycerin group(P〈0. 01 ) ; Bcl-2 proteins expression was significantly enhanced(P〈0. 05) ; Fas-L proteins expression was also lowed significantly(P〈0. 05). CONCLUSION Nitroglycerin could reduce the degrees of myocardial ceils apoptosis during continuous ischemia.
