摘  要:目的:探讨苦参碱对腹膜透析大鼠腹膜纤维化的影响和可能的机制。方法:采用高糖透析液+脂多糖复制腹膜纤维化模型。将大鼠随机分为对照组、模型组、苦参碱低剂量组和苦参碱高剂量组。治疗组在造模同时每日腹腔内注射不同剂量的苦参碱。35d后处死各组大鼠,行2h腹膜平衡试验,测量超滤量,检测腹膜功能;留取腹膜组织行病理切片做HE及Masson染色观察腹膜组织学改变;用RT-PCR法测定腹膜血管内皮生长因子(VEGF)和纤维连接蛋白(FN)表达率。结果:苦参碱治疗组的腹膜厚度和血管增生程度比模型组明显减轻,VEGF和FN的合成水平也明显下降。结论:苦参碱可以有效的防治腹膜纤维化的进程。OBJECTIVE To investigate the effects and possible mechanism of action of matrine for the treatment of experimental peritoneal fibrosis in rats. METHODS The peritoneal fibrosis rat model was induced by high glucose dialysate and lipopolysaccharide (LPS). The rats were randomly divided into four groups:control group, model group, low dose matrine treating group and high dose matrine treating group. The matrine treating group received daily intraperitoneat injection of different dose matrine, Thirty-five days later all rats were sacrificed. The peritoneal membrane function was investigated by peritoneal transport test; the abdominal membrane was stained with HE and Masson method, the levels of VEGF and fibronectin (FN) in peritoneum were detected by RT-PCR method. RESULTS The peritoneum of model group was thicker than that of matrine treating group. And the number of vascularity was higher than that of Matrine treating group. The concentration of VEGF and FN in blood and dialysate also decreased obviously in matrine treating group; The levels of VEGF and FIN in peritoneum in matrine treating group were significantly lower than those in model group. CONCLUSION Matrine are beneficial to the prevention and treatment of peritoneal fibrosis in experimental model rats.
