摘  要:为探讨温阳益气药的抗衰老机制,本文主要观察了老年大鼠脑M受体、心肌β受体、血浆环核苷酸、LPO、SOD、T_3、T_4皮质醇的改变以及黄芪的调节作用,分别用放射自显影方法、放射配基结合分析法、放射免疫分析法测定脑M胆碱受体、心肌β受体、血浆环核苷酸。结果表明:M受体包括M_1亚型受体主要分布在大脑皮质、海马、纹状体部位,老年大鼠这三个部位的密度显著低于青年大鼠,老年服黄芪组三个部位的受体密度显著高于老年对照组;老年大鼠心肌β受体最大结合容量Bmax显著低于青年大鼠,黄芪可以显著提高老年大鼠心肌β受体Bmax;老年大鼠cAMP,cGMP含量均显著升高,但cAMP/cGMP比值降低,黄芪可以纠正这种失衡。另外,老年大鼠血浆SOD活力降低,LPO含量升高,甲状腺激素T_3,T_4和皮质醇含量均显著降低,黄芪对这些改变均具有调节作用。说明:黄芪抗衰老的作用是多方面的,是针对多个系统的综合性调节。In order to study the and - aging mechanism of Wanning - yang and lonifyins - qi Chinese herbs, the change of brain muscarinic cholinergic receptors, heart (3 - adrenoceptor, plasma cyclic tuicleotides, LFO, SOI),T3,T4, cortisone of aged rats and the regulatory action of Astragalus menbranaccns were observed. An autoradiographic technique with 3H - QNB (quinuclidinyl benzilate) and 3H - PZ(pireri7.ip-ine) as radioactive probes was used to study the change of M - receptor and Ml subtype receptor densities in brain slices. It was found that the specific binding was mainly localized in cerebral cortex, hippocampus and stviatum. The mean grey of the specific binding of the above three brain areas of aged rats were significantly lower than those of young rats. The decrease of M - receptor density was about 24.81% , 14. \27r and 12.76% ( P < 0.05) respectively. The decrease of Ml subtype receptor density was about 15.87% , 8.69%, 12.36% (P < 0.05) .Parallel experiments revealed that when aged rats were treated by administration of Astragalus menbran aceus , the M - receptor and M L subtype receptor densities of cortex , hippocampus and striatum were significantly higher than untreated aged rats. The increase of M - receptor was24.15%,14.38%,13.47%(P<0.05), and of Ml subtype receptor, it was 16.63% ,9.81 %, 10. 32%(P<0.05) respectively. Radioligand binding assay revealed that the maximum binding capacity (Bmax) of aged rats was significantly lower than that of young rats, Astragalus menbran aceus took an obvious upregulatoiy action (P<0.05). The plasma cyclic nucleotides contents of rats were determined with radioimmunoassay (IUA), The results revealed that cAMP and cGMP contents of old rats were both significantly higher than that of young controls, while cAMP/cGMP ratio was significantly lower( P < 0. 05) .
