摘  要:目的用BDA(Biotinylated Dextran Amine)示踪大鼠皮质脊髓束,观察其在脊髓内的走行.方法采用成年SD大鼠10只,分A、B两组.A组用10%BDA注射到双侧皮质运动区,B组注射到左侧皮质运动区.3周后灌注取材,取C1~3、T3~5和T9~11节段脊髓做冰冻切片,行DAB染色.结果(1)横向观察:A组在双侧脊髓白质的后索以及前正中裂两侧观察到棕色阳性纤维.B组在右侧脊髓后索和左侧前正中裂观察到棕色阳性纤维;两组都有显色纤维从后索发出进入灰质且在灰质内分布均匀;(2)纵向观察:阳性纤维自吻端向尾端逐渐稀疏.结论BDA示踪技术可以直观地显示大鼠皮质脊髓束在脊髓内走行,阳性染色纤维的数量自吻端向尾端逐渐稀疏.Objective To trace and observe corticospinal tract coursing in rat's spinal cord with biotinylated dextran amine (BDA). Methods Ten adult Sprague-dawley rats were randomly divided into two groups:10 % BDA were stereotactieally injected into the both cortical motor area in group A and into left cortical motor area in group B. Three weeks later, animals were sacrificed. The spinal cord of C1-3, T3-5 and T9-11 were sliced and observed by DAB staining. Results (1) Horizontal survey: In group A, the BDA -labeled corticospinal tract fibers distributed in the whole posterior funiculus and in both banks of the anterior median fissure; In group B, the BDA - labeled fibers distributed in the right half of posterior funiculus and in the left bank of the anterior median fissure; In both groups, the BDA - labeled fibers, from posterior funiculus, entered grey matter and distributed in the whole grey matter; (2) Longitudinal survey:BDA - labeled fibers were gradually sparse from rostral side to caudal end. Conclusion BDA tracing Can clearly reveal the course of the corticospinal tract in rat' s spinal cord in which BDA - labeled fibers are gradually sparse from rostral side to caudal end.
