摘 要:目的:观察抑制性寡脱氧核苷酸对小鼠脾脏CD4+T细胞Th1分化的影响。方法:分离小鼠脾脏淋巴细胞,以免疫磁珠阳性选择法纯化CD4+T细胞,以荧光活化细胞分拣术分析其分选纯度,在抗-CD3ε抗体、抗-CD28抗体和IL-12作用下,加入抑制性寡脱氧核苷酸或对照寡脱氧核苷酸培养72 h后,用ELISA检测上清IFN-γ和IL-4的分泌,用RT-PCR检测细胞T-bet mRNA的表达。结果:抑制性寡脱氧核苷酸能明显抑制CD4+T细胞IFN-γ的分泌,促进IL-4的分泌,同时还抑制T-bet mRNA的表达。结论:在促Th1细胞因子刺激下,抑制性寡脱氧核苷酸对小鼠体外CD4+T细胞的功能性分化有作用,既促进Th2分化,又抑制Th1分化。Abstract: Objective To exlpore the effect of suppressive oligodeoxynucleotides ( Sup ODN ) on the Th 1 differentiation of CD4^+T splenetic lymphocytes in mice. Methods The splenetic lymphocytes of BALB/c mice were separated, and then CD4^+ cells were purified with immune magnetic CD4^+ microbeads (positive selection ). The purification was examined by fluorescence-activated cell sorter. CD4^+ cells, anti-CD3ε, anti-CD28, IL-12 and Sup ODN or control oligodeoxynucleotides (Con ODN ) were co-incubated for 72 h. IFN-γ and IL-4 in the supernatant were detected using enzyme-linked immunosorbent assay. The expression of T-bet mRNA in CD4^+ cells was tested by reverse transcription polymerase chain reaction. Results Sup ODN could significantly inhibit the re- lease of INF-γ and increase IL-4 production respectively (P 〈0.01 ). T-bet mRNA of CD4^+ lymphocytes was remarkably inhibited by Sup ODN as well ( P〉0.01 ). Conclusion In the presence of pro-Th1-cytokines, Sup ODN may affect the differentiation of CD4^+ T lymphocytes in vitro. Sup ODN can promote CD4^+ T cells to differentiate into Th2, and suppress them into Th1.
