摘 要:目的:检测人β-防御素(humanβ-defensin-1,hBD-1和humanβ-defensin-2,hBD-2)mRNA在复发性鼻息肉中的表达,分析hBD-1和hBD-2在复发性鼻息肉发病中作用。方法:应用逆转录-聚合酶链反应技术(reverse transcripatase-polymerase chain reaction,RT-PCR)检测鼻息肉患者术前鼻息肉、复发性鼻息肉患者手术后复发鼻息肉、鼻息肉患者术后恢复正常的鼻黏膜及正常对照鼻黏膜各10例中hBD-1和hBD-2 mRNA的表达。结果:hBD-1 mRNA在鼻息肉患者术前鼻息肉、术后复发鼻息肉、鼻息肉患者术后恢复正常鼻黏膜、正常鼻黏膜中均有表达,且组间差异无统计学意义(P>0.05);hBD-2 mRNA在鼻息肉术前组织和复发性鼻息肉组织中均有表达,在正常对照鼻黏膜和鼻息肉术后恢复鼻黏膜的中几乎没有表达。结论:hBD-1为固有表达,hBD-2为诱导表达。β-防御素可能在鼻息肉的发病中起一定的作用。Objective To observe the expression of human β- defensin-1 ( hBD - 1 ) and human β - defensin-2 ( hBD - 2 ) in recurrent nasal polyps, and to investigate the role of β- defensin in the recurrent nasal polyps. Methods Tissues of nasal polyps was obtained from 10 patients with nasal polyps undergoing endoscopic sinus surgery, recurrent nasal polyps from 10 patients 6 months after surgery, nasal mucosa from 10 recovered patients with nasal polyps postoperatively and,10 control subjects, hBD -1 mRNA and hBD -2 mRNA levels of tissue specimens in all groups were detected by reverse transcripatase-polymerase chain reaction (RT-PCR). Results There was no significant difference in hBD -1 mRNA level between the 4 groups ( P 〉0.05 ). Expression of hBD -2 mRNA was detected in patients with nasal polyps and recurrent nasal polyps, but rare in the recovered patients and the control subjects. Conclusion hBD- 1 is a constitutive expression and hBD-2 is an induced expression. β-Defensin may play an important role in forming the nasal polyps.
