摘  要:目的探讨益智通脉颗粒对血管性痴呆大鼠脑组织VEGF表达的影响。方法健康成年雄性Wister大鼠50只随机分为正常对照组、模型对照组、中药大剂量组、中药小剂量组、尼莫地平组各10只,采用双血管阻断法制备拟血管痴呆大鼠模型。术后7d模型组及药物组分别给药,连续给药60d。采用免疫组化法观察脑组织VEGF的表达。结果中药大、小剂量组与模型组及尼莫地平组比较有显著差异;中药大、小剂量组之间呈现正相关量-效关系。结论益智通脉颗粒可通过上调VEGF表达,促进内皮细胞增殖及毛细血管新生,从而起到脑保护作用。Objective: To observe the effects of Yizhitongmai Granule on VEGF of cerebral tissue of vascular dementia(VD)model rats. Methods: 50 male Wistar rats were randomly divided into 5 groups: the normal group,the model group, high - dose group (treated with high dose Yizhitongmai Granule), low - dose group and nimodipine group. Rat VD model was established by the method of both of vessel -blocked, and then 7d later, treating the rats with medicine for 60d. SP immunohistochemical staining was used to examine expressions of VEGF of cerebral tissue. Results: There were significantly difference by high -dose and low- dose group comparing with model group and nimodipine group. There were positive correlation between high-dose and low-dose group. Conclusion: Yizhitongmai Granule can facilitate proliferation of endothelial cell, accelerate vascularization and increase vasucular permeability by upregulate VEGF expression to protection brain.
