摘  要:目的:研究高压氧治疗对大鼠颅脑外伤后神经元凋亡的影响,以探讨高压氧治疗颅脑外伤的机制。方法:(1)运用RT-PCR技术,检测高压氧治疗后大鼠脑组织Bcl-2、Bax、Caspase-3基因的变化。(2)运用TUNEL染色法,检测高压氧治疗对TUNEL染色阳性细胞的影响。结果:(1)大鼠颅脑外伤后6小时起高压氧治疗后,与对照组相比Bcl-2基因表达增加,而Bax基因表达下降,Bcl-2/Bax比值明显上升,Caspase-3基因表达增加,均具有统计学意义。(2)高压氧治疗后与对照组相比TUNEL染色阳性细胞数明显减少。结论:高压氧治疗对大鼠颅脑外伤后神经元的凋亡具有抑制作用,从而减轻大鼠颅脑外伤后的继发性脑损害。Objective: To make a study of the inhibited effect of hyperbaric oxygenation on apoptosis of nerve cells following the rat's craniocerebral trauma, and to probe into the mechanism of using hyperbaric oxygenation to cure the craniocerebral trauma.Methods: (1)Use RT-PCR technique to observe the changes of Bcl-2, Bax and Caspase-3 genes in the rat's brain tissue after hyperbaric oxygenation. (2)To take advantage of TUNEL staining method to observe the effect of hyperbaric oxygenation on the positive cells dyed by TUNEL staining method.Results: (1)Through the hyperbaric oxygenation six hours after the rat's craniocerebral trauma, compared with the control group, Bcl-2 gene expression increased, but Bax gene expression declined, the ratio of Bcl-2 to Bax rose noticeably, Caspase-3 gene expression increased obviously, and all of them were of statistic significance. (2)Compared with the control group, after the hyperbaric oxygenation, the positive cells dyed by TUNEI declined noticeably.Conclnsions: Hyperbaric oxygenation has inhibited effect on apoptosis of nerve cells following the rat's eraniocerebral trauma, which can alleviate the secondary lesion following the rat's craniocerebral trauma.
