摘 要:通过PCR-SSCP技术检测了大通牦牛IGF-I基因的遗传多态性,对具有SSCP多态性的片段进行克隆和测序,找出等位基因的多态位点,将该位点多态性与大通牦牛生长性状进行相关性分析.结果显示:PCR-SSCP检测到大通牦牛具有AA、AB、BB 3种基因型,A(B)等位基因频率分别为0.902 8(0.097 2);对IGF-I基因第1内含子进行序列比对,发现一处单碱基C的缺失/插入,造成了该位点多态性的出现;6月龄和12月龄基因型为AA的大通牦牛的体质量和体斜长最小二乘均值显著高于AB和BB基因型(P<0.05);18月龄基因型为AB的大通牦牛的体高最小二乘均值显著高于BB型(P<0.05),说明大通牦牛品种中基因型为AA的个体生长性状有高于AB和BB基因型的趋势,可以作为大通牦牛品种选育的目标基因型.The polymorphism of IGF-Ⅰ gene of Datong yak was detected by using PCR-SSCP technology,and the PCR products with polymorphism fragments were cloned and sequenced to detect single nucleotide polymorphism;the relationship between the polymorphism and Datong yak's growth traits was analyzed. Results indicated that the frequences of allele genes A(B) is 0. 902 8(0. 097 2) in Datong yaks;the Cbase-deletion in intronl of IGF-Ⅰ gene was detected by sequence align, causing polymorphism in Datong yaks;the body weight and body length of individuals with genotype AA at 6 and 12 months of age were signifieantly higher than those with genotype BB,AB(P〈0.05);the body height of individuals with genotype AB at 18 months of age were significantly higher than those with genotype BB(P〈0. 05). The growth traits of individuals with genotype AA were significantly higher than those with genotype BB,AB in Datong yaks. This genotype could be selected as target-genotype in Datong yak.
