山药多糖对小鼠CCL_4肝损伤的保护作用

摘　　要：目的:探讨山药多糖对小鼠CCL4肝损伤的保护作用及可能作用机制。方法:昆明种小鼠50只随机分空白对照组;模型组;高、中、低山药多糖组,每天分别给予50 mg/kg、100 mg/kg、200 mg/kg剂量的山药多糖灌胃,连续8 d,末次灌胃2 h,空白对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%CCL4调和油溶液,24 h眼球取血测定血清中ALT、AST活性和MDA含量。取肝脏,称重计算肝体指数;制备肝匀浆,按试剂盒要求测定MDA、SOD、GSH的含量。结果:山药多糖可减轻实验性肝损伤所致炎性反应,降低肝体指数,降低血清中ALT、AST活性(P(0.05)和MDA的含量(P(0.01);提高肝细胞中GSH的含量(P(0.01)及降低MDA的含量(P(0.01)。结论:山药多糖对CCL4造成的实验性肝损伤有保护作用,其机制可能与山药多糖抗氧化作用有关。Objective To explore the protective effect of Yam polysaccharide on CCL4-induced liver injury. Methods Kunming mice were randomly divided into control group, pathological model group and low, medium, high dosage of Yam polysaccharide groups. Three Yam polysaccharide subgroups were given Yam for 8 days by the body weight dosage 50 mg/kg, 100mg/kg,200mg/kg, respectively. The control group was intra-peritoneally injected blend oil solution, and the other groups was intra-peritoneally given 0.15% CCL4 mixed with blend oil solution at 2h after last adminstration. The activity of ALT and AST,the content of MDA in blood serum were all determined after 24 hours. The liver index was calculated and the contents of MDA,SOD, GSH in liver homogenate were determined. Results Yam polysaceharide could alleviate the inflammation in response to experimental liver injury, decreased the liver index, ALT and AST activity（ P 〈 0.05 ） and the content of MDA （P 〈0.01 ） in serum. In addition, it also increased the content of GSH （P 〈 0.01 ） and decreased the content of MDA （P 〈 0.01 ） in the liver. ConclusionYam polysaccharides can protect the CCLn-induced experimental liver injury, and its mechanism might be related to antioxidant activity of yam polysaccharides.
