摘 要:以盐地碱蓬茎尖为外植体,探讨了不同激素浓度配比对植株再生的影响.结果表明,以盐地碱蓬茎尖为外植体,在附加1.0mg/L6-BA和0.5mg/LIAA的MS培养基上培养30天可诱导出不定芽,不定芽的诱导频率达到100以上,每个外植体的平均出芽数达到3.63,60天后形成丛生芽.不定芽转至MS培养基中培养10天生根形成完整植株.本文建立了真盐生植物盐地碱蓬高效的再生体系,为遗传转化体系的创建奠定了基础.Meristem tip as explants of euhalophyte Suaeda salsa was used to examine effect of different combinations of growth regulators on the plant regeneration. The results showed that the best combination for the induction of adventitious shoots was MS medium supplemented with 1.0 mg/L 6 - benzylaminopurine (6 - BA) and 0.5 mg/L indole - 3 - acetic acid (IAA), and the induction frequency was up to 100%. Under the most favorable condition, one explant could regenerate 3.63 adventitious shoots after 30 days and formed cluster shoots after 60 days. Rooting was achieved on MS medium without any plant growth regulators after adventitious shoot was transplanted for 10 days. An efficient plant regeneration protocol in vitro for euhalophyte S. salsa was achieved via organogenesis from meristem tip explants and laid the foundation of its efficient transformation system.
