摘 要:目的探讨芍药苷(Pae)通过免疫性肝损伤大鼠枯否细胞(KC)影响肝星状细胞(HSC)的增殖能力。方法用卡介苗加脂多糖诱导大鼠肝损伤模型,肝脏原位灌流分离大鼠KC和HSC,观察共培养的形态学改变,观察Pae作用后的KC培养上清对HSC增殖的影响。结果免疫性肝损伤大鼠KC能明显促进HSC的增殖,在Pae作用后,KC培养上清能抑制HSC的增殖。结论KC是Pae发挥作用的靶细胞之一,Pae可通过作用于KC抑制HSC的增殖。Objective To investigate the effects of kupffer cell(KC) culture medium on the proliferation of hepatic stellate cells (HSC) isolated from rats with immunological liver injury and the role of paeoniflorin. Methods Rats were induced by Bacillus Calmette-Guerin plus lipopolysaccharide, KC and HSC were isolated by means of liver perfusion in suit. The morphology changes in co-cluture were observed on the 7th day. The effects of KC culture medium supplemented with different concentrations of paeoniflorin on HSC proliferation were detected by MTT assay. Results KC supernatants could promote the HSC proliferation. KC supernatants treated with paeoniflorin could decrease the promoting effects of KC on HSC proliferation. Conclusion Paeoniflorin can decrease effects of KC on promoting HSC proliferation via modulation for KC function.
