摘 要:目的研究模拟微重力对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC)形态及微丝骨架的影响。方法利用回转器模拟微重力,将原代培养的HUVEC分为回转组和静止对照组,培养72 h,相差显微镜观察两组细胞的形态;电镜观察微丝(microfilament,MF)的形态分布。结果静止培养的细胞贴壁良好,形态规则,微丝在细胞核及细胞膜的周围多呈束状分布,回转组的细胞贴壁不好且微丝排列紊乱、微丝变短,微丝松散、稀少而且不成束排列。结论模拟微重力情况下细胞形态有所改变,微丝骨架分布的有序性降低。Objective To investigate the effect of simulated microgavity on microfilament and morphology in primary culture of human umbilical vein endothelial cells ( HUVEC ). Methods Primary culture of HUVEC was exposed to simulated microgravity condition and stationary condition for 72 h. Electron microscope and inversion microscope were used to observe the microfilaments and morphology of HUVEC . Re- sults The morphology and ultrastructure of HUVEC cultured under microgravity changed. Microfilament distributed in disorder in simulated microgravity group, and became shorter compared with stationary control group, mierofilament highly disorganized and lost its array. Conclusion The morphology of cells and the distribution of filaments may change under simulated microgravity.
