摘  要:目的检测转录因子Ets-1在大鼠动情周期卵巢组织中的表达规律,探讨其在卵泡发育、黄体形成及退化中的作用。方法分别在动情周期的不同时期收集大鼠卵巢标本,采用逆转录聚合酶链反应(RT-PCR)和免疫组化的方法观察Ets-1 mRNA在大鼠动情周期各不同时期表达水平的差异,以及Ets-1蛋白在大鼠动情周期卵巢组织中的表达分布情况。结果RT-PCR结果显示,Ets-1 mRNA在大鼠动情周期各不同时期均有表达,卵泡发育过程中Ets-1 mRNA表达逐渐增强,在动情前期表达最强,动情后期最弱;免疫组化结果显示,Ets-1蛋白在卵巢各级卵泡和黄体中表达,主要表达于卵母细胞、颗粒细胞和黄体细胞。结论Ets-1可能在大鼠动情周期中的卵泡发育、黄体生成及退化过程中起着重要的调节作用。Objective To examine the spatiotemporal expression pattern of transcription factor Ets-1 in rat ovary and investigate its function in ovarian follicular development and corpus luteum (CL) formation and degeneration during the estrous cycle. Methods Rat ovaries were collected at diestrus, proestrus, estrus and metestrns, respectively. Total RNA was extracted and reverse transcription polymerase chain reaction (RT-PCR) was performed to observe the expression of Ets-1 mRNA in rat ovary at different stages during the estrous cycle. Paraffin-embedded tissue sections were prepared and immunohistochemistry was used to demonstrate the localization of Ets-1 protein in rat ovary during the estrous cycle. Results RTPCR results showed that the expression of Ets-1 mRNA increased coinciding with the development of the dominant follicle,which was strongest at proestrus and weakest at metestrus. Immunohistochemical analysis demonstrated the expression of Ets-1 protein in follicles of different sizes and in CL at different stages during the estrous cycle, and the immunostaining was predominantly localized in oocytes, granulosa cells and luteal cells. Conclusion Ets-1 may play important roles in the regulation of follicular development and luteal function during the estrous cycle.
