摘　　要：目的：探讨转化生长因子（TGF）-β1在伤寒沙门菌诱导巨噬细胞凋亡过程中的调节作用。方法：ELISA检测伤寒沙门菌诱导小鼠巨噬细胞系RAW264.7培养上清中TGF-β1分泌水平,实时定量PCR检测伤寒沙门菌诱导小鼠巨噬细胞TGF-β1mRNA的表达,流式细胞术观察外源性TGF-β1对伤寒沙门菌诱导巨噬细胞凋亡的影响。结果：伤寒沙门菌感染组巨噬细胞TGF-β1的分泌和mRNA的表达水平显著高于对照组（P〈0.05）;伤寒沙门菌热杀死组、伤寒沙门菌感染组巨噬细胞凋亡率显著高于对照组（P〈0.05）,LPS＋伤寒沙门菌感染组的巨噬细胞凋亡率显著高于细菌脂多糖（LPS）组（P〈0.05）;1～100μg/L3个浓度的TGF-β1组的巨噬细胞凋亡率显著低于伤寒沙门菌感染组（P〈0.05）,并呈一定的剂量依赖关系。结论：伤寒沙门菌能够诱导小鼠巨噬细胞系RAW264.7表达TGF-β1,不同剂量的外源性TGF-β1能够抑制伤寒沙门菌诱导巨噬细胞凋亡的过程,提示TGF-β1在伤寒沙门菌诱导巨噬细胞的凋亡过程具有调节作用。Objective： To explore the effect of TGF-β1 on apoptosis of murine macrophages RAW 264.7 induced by Salmonella typhimurium strain. Methods： The secretion of TGF-β1 in the culture supernatant of macrophages was examined using ELISA. The expression of TGF-β1 mRNA in the macrophages was analyzed by real time PCR. The apoptosis of macrophages was determined by FACS. Results： The secretion of TGF-β1 and the expression of mRNA in the group of Salmonella Spp. were markedly promoted than that in control group （P 〈 0.05）. The percentage of apoptosis in the group of Salmonella Spp. Killed and Salmonella Spp. was significantly increased than that in control group （P 〈 0.05）, and the percentage of apoptosis in Salmonella Spp. with LPS group was also significantly higher than that in LPS group （P 〈 0.05）. The percentage of apoptosis in the Salmonella Spp. with 1 -100 I.Lg/L TGF-β1 group was remarkable decreased than that in Salmonella Spp. group in a dose-dependent manner （P 〈 0.05）. Conclusion： TGF-β1 can be expressed in the process of apoptosis of macrophages infected by Salmonella typhimurium strain, and TGF-β1 at different dosage can inhibit apoptosis of macrophages. TGF-β1may regulate the apoptosis of macrophages infected by Salmonella typhimurium strain.
