摘 要:目的观察维生素A酸(RA)对体外培养小鼠囊胚细胞毒性作用。方法获取妊娠3.5d小鼠囊胚,分别在含0、1和10μmol/LRA的M199培养液中培养24h,用荧光染料Propidium(PI)和Bisbenzimide(Hoechst 33258)特异性染色,分别计数囊胚内细胞总数以及内细胞群(ICM)和滋养层(TE)细胞数。结果RA可导致小鼠囊胚内细胞总数以及ICM和TE细胞数减少。结论RA对小鼠早期胚胎的发育具有细胞毒性作用,从而影响早期胚胎的生长发育。Objective To examine the cytotoxic effect of retinoic acid on mouse blastocysts cultured in vitro. Methods Murine 3.5-day blastocysts were exposed to doses of 0, 1 and 10 μmol/L retinoic acid for 24 h, and stained with fluoroehrome propidium iodide (PI) and bibenzimide (Hoechst 33258). The number of total cells in blastoeysts and the inner cell mass (ICM) and trophectoderm (TE) cells was counted separately. Results Retinoic acid could result in a significant reduction in the number of total cells in hlastoeysts and the trophectoderm/inner cell mass lineage in mice. Conclusion Retinoic acid had eytotoxie effects against the development of early embryos in mice, which could interfere with the growth and development of early embryos.
