摘 要:本研究测定了不同培养基、碳源、氮源、温度、pH条件下臭椿盘长孢刺盘孢(Colletotrichum gloeosporioides Penz.)菌株SQD-107的生长、产孢和孢子萌发情况。该菌株在PSA培养基上生长最快、产孢最多;在供测碳源中病原菌对蔗糖、葡萄糖利用较好,供测氮源中对硝酸铵、氯化铵利用较好。菌丝生长的适宜温度范围为10~35℃,最适25℃;产生分生孢子的适宜温度范围为10~30℃,最适25~30℃;孢子萌发的适宜温度范围为5~35℃,最适25℃。在pH3~10的范围内该菌均能生长和产孢,菌丝生长最适pH7~8;产生分生孢子最适pH为7,孢子萌发最适pH6~8。孢子的致死温度为60℃,持续时间10min。In our previous study, the SQD- 107 was isolated from anthracnosed leaves of tree- of- heaven (Ailanthus altissima) in China and was morphologically identified as Colletotrichum gloeosporioides Penz. In the present study, the effect of media, carbon sources, nitrogen sources, temperatures and pH values on mycelial - growth, sporulation and conidial germination of SQD - 107 was tested and analyzed. The results indicated that potato sucrose agar (PSA) was found the best medium for mycelial growth and sporulation of the fungus. The mycelial grew much faster and produced significantly more conidia on PSA. When different C and N sources were tested, sucrose or glucose and NH4 NO3 or NH4 Cl were the best sources for the fungal mycelial SQD - 107 could grow normally at 10 - 35 ℃, but the best mycelial growth was recorded at 25 ℃. The growth. suitable pH values were from pH3 to pH10, with the optimum being pH7 - 8. SQD - 107 sporulated normally at 10 - 35 ℃ and pH3 - 10, with the optimums being 25 ℃ and pH7, respectively. The apt temperatures for conidial germination ranged from 5 ℃ to 35 ℃, and the optimum was 25 ℃. The range of pH values for conidia germination was pH2 - 11, the optimum being pH6 - 8. Conidia of the fungus were inactivated at 60 ℃ for 10 min.
