摘  要:目的:建立检测中药材淡竹叶中的总黄酮(以芦丁为外标)和3种黄酮苷(异荭草素、牡荆苷、苜蓿素7-0葡萄糖苷)的有效、精确而可靠的HPLC/DAD方法。方法:色谱柱为Agilent SB—C18柱(250mm×4.6mm,5.0μm),用乙腈和0.5%的冰醋酸为流动相梯度洗脱,波长为350nm。结果:其加样回收率为97.1%-102.3%,芦丁和3种黄酮苷均得到满意的分辨率和线性范围,同时为了确保方法的有效性,还分析了不同产地淡竹叶中的总黄酮和3种黄酮苷的含量。结论:HPLC检测方法可用于淡竹叶及其相关产品的质量控制。AIM: To develop an effective, accurate and reliable HPLC/DAD method for determination of total flavonoids (rutin served as external standard) and three flavonoid glycosides (isoorientin, vitexin and triein 7-0- glucoside) in traditional Chinese herb Lophatherum gracile Brongn. METHODS: The chromatographic separation was performed on an Agilent SB-C18 column (250 mm ×4.6 ram, 5.0 μm) with a gradient elution programme using the mixture of acetonitrile and 0.5% aqueous acetic acid (v/v) as mobile phase at 350 nm. RESULTS: The recovery of the method was in the range of 97.1%-102.3% , and the four compounds showed good linear relation ( R2 〉 0. 999) in a relatively wide concentration range. In addition, the content of total flavonoids and three flavonoid glycosides in L. gracile growing in different locations of China was determined to establish the effectiveness of the method. CONCLUSION: The developed HPLC assay could be readily utilized as a quality control method for L. gracile and its related medicinal preparations.
