Plaque Cloning and Virulence Determination of Vaccine Strain for Newcastle Disease Virus and Isolated Strains

摘  要:2株NDV贵州分离株(GN、ND99)与2株疫苗毒株(Ⅰ系和Ⅱ系)经检测为生物异质性毒株。利用蚀斑克隆技术对以上4株毒株进行蚀斑纯化,结果得到11株克隆毒株。其中,10株克隆毒株在鸡胚成纤维细胞上形成圆形、透明无色蚀斑,1株形成圆形、透明、红色蚀斑。经RT-PCR检测,11株克隆毒株中6株为弱毒,形成蚀斑的大小在0.4~0.8mm之间;5株为强毒,形成蚀斑的大小在1.0~2.0mm之间。结果表明:所形成蚀斑的大小与病毒毒力具有一定的相关性。Two Newcastle disease virus(NDV) isolates from Guizhou province(GN,ND99)and two vaccine strains( Ⅰ , Ⅱline) were determined as the heterogeneous viruses of biological property. 11 cloning strains havd been required by plaque cloning technique. 10 cloning strains could form round,transparent and colorless plaques on the monolayer culture of chicken embryo fibroblast cells(CEF),the other strain could form round,transparent and red plaque. The results of RT-PCR detection showed that 6 cloning strains belonged to the mesogenic viruses with the plague size of 0.4 to 0.8 mm,5 cloning strains belonged to the velogenic viruses with the plague size of 1.0 to 2.0 ram. It demonstrated that some correlation was found between the size of plaque and virulence of NDV.
