摘 要:应用键合型3,5-二甲基苯基氨基甲酸酯淀粉类手性固定相色谱柱(Chiralpak IA),在正相系统的高效液相色谱条件下首次直接拆分了具有抗病毒活性的一系列含氟烷氧取代α-氨基膦酸酯。在选定的条件下,8种α-氨基膦酸酯均可达到基线分离(Rs>2.15)。同时,从保留因子(k′)、分离因子(α)和分离度(Rs)等方面考察了对映体保留和拆分的影响因素,并制备了一对对映异构体。研究结果表明,化合物结构和流动相组成对化合物外消旋体分离具有重要影响。By using immobilized amylose-tris (3, 5-dimethyl- phenylcarbamate) CSP (Chiralpak IA), the racemie mixture of a series of α-aminophosphonates was resolved in the normal phase mode. The chromatographic parameters such as the retention factor (k') ,the separation factor ( a ), and the resolution (Rs) of the solutes were studied. The factors influencing the ehiral separation were also developed. A reasonably good baseline separation for these compounds was achieved using Chiralpak IA column with Rs 〉 2.15. The optimum condition for separating and preparing a pair of enanfiomers was investigated. It was found that both the compound structure and the composition of mobile phase had significant effects on their chromatographic behaviors.
