摘  要:目的观察动脉粥样硬化(AS)兔模型中热休克蛋白60(HSP60)的表达及其与斑块稳定性之间的关系和普罗布可对HSP60表达的影响。方法20只新西兰大白兔随机分正常对照组(n=6)、高脂饮食组(n=8)、普罗布可组(n=6)。对照组予以普食喂养,高脂饮食组和普罗布可组予高脂喂养。4周后高脂饮食组和普罗布可组行髂动脉内膜球囊损伤术。术后普罗布可组每只兔子加用普罗布可1000mg/d口服,10周末处死动物。采用HE染色观察动脉病理形态学变化,免疫组化染色检测动脉中HSP60的表达,酶联免疫吸附法测血清中的C反应蛋白(CRP)、白细胞介素(IL)-6、氧化低密度脂蛋白(ox-LDL)、HSP60的水平。结果高脂饮食组动脉可见典型的动脉粥样硬化斑块,普罗布可组无典型斑块形成,内膜增厚较为明显;与高脂饮食组相比较,普罗布可组血清CRP、IL-6、ox-LDL、HSP60水平降低;普罗布可组内膜和中膜HSP60的表达较高脂饮食组明显减少。血清HSP60水平与CRP、IL-6、ox-LDL水平正相关。结论HSP60在兔AS模型中的表达水平明显增加,与斑块的不稳定性有关,普罗布可可抑制兔AS模型血清和斑块中HSP60的表达。Objective To investigate the expression of heat shock protein 60 (HSP60) and its association with the stability of plaque on the atherosclerosis rabbit model, and the influence of probucol on the expression of HSP60. Methods Twenty New Zealand rabbits were randomly divided into control group, high-cholesterol group and probueol group. The control group was raised with normal food and the 2 other groups were raised with high-cholesterol food. After 4 weeks, the iliae artery lesion was induced by balloon angioplasty in the rabbits of the high-cholesterol group and the probueol group. Then 1000 mg probucol was added to each rabbit's daily food in probucol group. Ten weeks later all rabbits were sacrificed. The expression of HSP60 in the iliae artery lesion was determined by immunohistochemical assay. Enzyme linked immunosorbent assay were used to detect serum HSP60, C reactive protein (CRP), interleukin (IL)-6 and ox-LDL-C levels. Results Atheromatous plaque was observed in high-cholesterol group, and only thicker tunica intima was observed in probueol group. Compared with baseline, the serum levels of HSP60, CRP, IL-6 and ox-LDL-C were decreased in probueol group compared with high-cholesterol group. The expression of HSP60 in the artery was lower in probueol group than in high-cholesterol group. The serum levels of HSP60 was positively correlated with the levels of CRP, IL-6 and ox-LDL-C. Conclusions The HSP60 level increases significantly in the atherosclerotic rabbits, which may has a correlation with the instability of the plaque. Probueol can significantly lower the HSP60 level in serum and the atherosclerotic plaque.
