摘  要:目的观察外源性内毒素脂多糖(lipopolysaccharide,LPS)作用于大鼠大脑皮层星形胶质细胞(astrocyte,AC)时,NO产生和释放的变化,以及选择性诱导型一氧化氮合酶(inducible nitricoxide synthase,iNOS)抑制剂S-甲基异硫脲(S-methylisothiourea,SMT)对其的影响。方法运用Griess重氮化反应法检测NO代谢产物NO2-/NO3-来反映正常组、LPS(10μg/mL)诱导后及加入SMT(1μmol/mL)的LPS诱导后NO的活性。结果LPS作用后,AC合成和分泌NO增加,呈时间依赖性,且在24 h增加最明显;SMT能明显抑制LPS诱导AC的NO过分泌(P<0.001)。结论在体外培养的AC,LPS可诱导NO的合成和释放增加,该过程主要是通过激活iNOS表达实现的。Objective To explore the change of nitric oxide (NO) by LPS in rat astrocytes, and the effect of SMT on it. Meth- ods NO was detected by Griess method in normal astrocytes, LPS (10 ~g/mL) induced astrocytes, SMT (1 umol/mL) added and LPS induced astrocytes. Results NO increased in a time - dependent manner when cells were induced by LPS, especially on 24 h, SMT inhibited the secretion of NO induced by LPS in astrocytes. Conclusions LPS could stimulate the release of NO, which could be down - regulated by SMT remarkably. As a result, we can say the main mechanism that contributes to the accretion of NO secretion might be the activation of iNOS.
