慢性排斥反应移植肾组织TIAF-1的表达及其意义

摘　　要：目的探讨移植肾慢性排斥反应(CR)时转化生长因子-β1诱导的抗凋亡因子-1(TIAF-1)的表达及其意义。方法采用SP免疫组织化学染色法对6例因肾细胞癌行肾切除的未受肿瘤侵犯的正常肾组织和8例供肾正常肾组织、16例急性排斥反应和28例慢性排斥反应移植肾组织中TIAF-1的表达进行观察,并对间质浸润细胞中CD3、CD20、CD68阳性细胞数进行分析。结果正常供肾组织未见TIAF-1表达。急性排斥反应移植肾间质TIAF-1阳性细胞数明显增加,肾小管上皮细胞呈微弱的TIAF-1表达。在慢性排斥反应移植肾组织中,TIAF-1在间质浸润细胞中的表达呈弥漫性、强阳性,较多的肾小管上皮细胞呈高度染色。移植肾间质浸润的TIAF-1阳性细胞数与CD3和CD20阳性细胞数呈正相关。结论TIAF-1在慢性排斥反应移植肾组织中的表达可能与Th1细胞向Th2细胞的免疫偏离有关,移植肾肾小管上皮细胞的TIAF-1表达可能反映了某种保护性机制的诱导。Objective To investigate the characteristics of TGF-β1-induced antiapoptotic factor-1（TIAF-1） expression in renal allograft of chronic rejection（CR） and to study the possible pathogenic role of TIAF-1 in chronic allograft rejection.Methods Immunohistochemistry of streptavidin-perosidase（SP） method was applied in the observation of the expression of TIAF-1 in each group,including tissues obtained from the unaffected part of nephrectomized kidneys with renal cell carcinoma（n=6） and cadaveric donor of normal kidneys（n=8） as the control group,tissues with acute rejection（AR,n=16） and those with chronic rejection（CR,n=28）,and in the analysis combined with the number of positive cells of CD3,CD20 and CD68 infiltrated in the interstitium.Results The expression of TIAF-1 was not detectable in normal kidney.The expression of TIAF-1 positive cells in the interstitium of renal allografts with acute rejection was up-regulated and TIAF-1 was weakly expressed in tubular epithelial cells.In tissues with chronic renal allograft rejection,a high and diffusive expression of TIAF-1 was found in the positive cells infiltrated in the intersitium of the renal allografts,with more tubular epithelial cells highly stained,most of which were CD3 and CD20 positive cells.Conclusion The expression of TIAF-1 in renal allografts with chronic rejection might be correlated with the immune deviation of Th1 to Th2 cells.The expression of TIAF-1 in tubular epithelial cells of renal allografts may be induced by a certain protective mechanism.
