摘  要:目的探讨激活过氧化物酶体增殖物激活受体α(peroxisome proliferators-activated receptorsα,PPARα)对海马神经元胆固醇外流的影响及可能机制。方法在大鼠海马神经元培养液中加入0.1mmol/L氯贝丁酯,用RT-PCR方法观察肝X受体(liver X receptor,LXR)及其靶基因ATP结合盒转运体A1(ATP binding cassette transporter A1,ABCA1)mRNA表达,用胆固醇氧化酶-比色法检测细胞胆固醇排出量的变化。结果氯贝丁酯降低海马神经元LXR和ABCA1mRNA表达(P<0.05),降低胆固醇排出量(P<0.05)。结论激活PPARα可能通过抑制LXR-ABCA1途径减少海马神经元胆固醇外流。Objective To investigate the effect of peroxisome proliferators-activated receptors a(PPARα) activation on cholesterol efflux in hippocampal neurons. Methods Hippocampal neurons were treated with 0. 1 mmol/L clofibrate and dissolved in dimethylsulfoxide for 48 h. The expression of liver X receptor (LXR) mRNA and its target gene ATP binding cassette transporter A1 ( ABCA1 ) mRNA were measured by RT-PCR. Cholesterol efflux of neuron was determinated by enzymatic-colorimetric assay. Results The expression of LXR mRNA and ABCA1 mRNA were reduced and cholesterol efflux was significantly decreased in clofibrate-treated hippocampal neurons. Conclusions PPARα activation may be through inhibiting the LXR-ABCA1 pathway to reduce cholesterol efflux in hippocampal neurons.
