摘 要:目的运用蛋白质组学技术观察益智健脑颗粒对SAMP8小鼠海马蛋白质表达的影响,从而探讨益智健脑颗粒治疗阿尔茨海默病(AD)的部分作用机制。方法将6月龄SAMP820只随机分为治疗组(n=10)和对照组(n=10),治疗组以益智健脑颗粒浓缩液灌胃,对照组以等剂量双蒸水灌胃。8w后,应用双向凝胶电泳(2-DE)技术分别分离治疗组和对照组SAMP8海马区组织总蛋白,经胶体考马斯亮蓝染色,利用ImageS-canner图像扫描仪及ImageMaster双向凝胶电泳软件对图像进行扫描分析,将有意义的差异蛋白质点经基质辅助激光解吸电离飞行时间质谱(MAL-DI-TOF-MS)分析得到肽质量指纹图谱(PMF),经MSDB和NCBI数据库查询,鉴定差异蛋白质点。结果鉴定出治疗组13个差异表达大于2倍的蛋白质,其中有8个上调,5个下调。结论益智健脑颗粒可调节SAMP8小鼠海马组织的多种蛋白质表达,提示该药具有多靶点治疗的作用。Objective To explore the remedial effect of Yizhijiannao-granule's probable mechanism on Alzheimer's disease (AD). Methods 6-month-old SAMP8 rats( n = 20) were randomly divided into treated group( n = 10) and control group( n = 10). Rats in treated group were treated with Yizhijiannao-granule and those in control group treated with double distilled water. Eight weeks later,total protein in hippocampus tissue were separated and identified. Results 13 Differentially expressed protein spots which were more than two folds were i- dentified, of which 8 protein spots were up-regulated and five protein spots were down-regulated. Conclusions Multiple hippocampus protein spots of SAMP8 mice can be regulated by Yizhijiannao-granule, showing Yizhijiannao-granule has a multi-target remedial mechanism of AD.
