摘 要:目的以核糖体DNA(rDNA)的第一与第二内转录间隔区序列(ITS-1及ITS-2)鉴定从广州动物园鸬鹚体内分离出来的鲁道夫对盲囊线虫种类。方法将鲁道夫对盲囊线虫样品GZ1、GZ2、GZ3和GZ4的ITS-1、5.8S、ITS-2进行PCR扩增及序列分析,并与GenBankTM公布的Contracaecum rudolphii A、B种相应序列进行比较。结果来自鸬鹚体内的4条鲁道夫对盲囊线虫具有相同长度的ITS序列,其ITS-1、5.8S、ITS-2分别为451、157和268bp,与GenBankTM公布的鲁道夫对盲囊线虫B种(C.rudolphii B)序列几乎完全一致,但在第108位缺失T。结论来自广州动物园鸬鹚体内的对盲囊线虫是C.rudolphii B。Objective To perform molecular identification of Contracaecum rudolphii (s.1.) isolated from Phalacrocorax carbosinensis by sequencing of the first and second internal transcribed spacers (ITS) of ribosomal DNA (rDNA). Methods Samples (GZ1-GZ4) were isolated from Phalacrocorax carbosinensis inhabiting in Guangzhou zoo. ITS-1, 5.8 S, and ITS-2 of rDNA were amplified by PCR and the amplicons were sequenced. The sequences were then compared with the sequences of C. rudolphii A and C. rudolphii B obtained from GenBankTM. Results GZ1-GZ4 had the same ITS sequences. The size of ITS-1, 5.8 S and ITS-2 was 451, 157 and 268 bp, respectively. The sequence of ITS was consistent with of the sequence of C. rudolphii B, with the exception of a deletion at the 108^th base in ITS. Conclusion Samples of C. rudolphii (s.1.) isolated from P. carbosinensis in Guangzhou zoo were identified as C. rudolphii B.
