肝细胞生长因子的血清浓度与胃癌进展的相关性

摘　　要：目的:检测胃癌患者血清中肝细胞生长因子(hepatocyte growth factor,HGF)水平,分析血清HGF水平与胃癌临床病理特征之间的关系,探讨胃癌患者血清中HGF表达的临床意义。方法:用酶联免疫吸附夹心法在术前检测80例胃癌患者血清HGF水平。结果:胃癌患者血清HGF水平(2.94±2.67ng·mL-1)明显高于正常对照组(1.02±0.31ng·mL-1)(P=0.002);分层分析发现,随着疾病进展,血清HGF水平逐渐升高(I期2.19±2.12ng·mL-1,II期2.53±2.38ng·mL-1,III期3.92±3.45ng·mL-1,IV期4.13±3.71ng·mL-1),Ⅲ期、Ⅳ期分别与I期进行比较,均有显著差异(P<0.05),有淋巴结及肝脏转移者的血清HGF水平升高更为明显,P值分别等于0.016和0.009。结论:胃癌患者血清HGF水平明显高于正常对照组,而且血清HGF的表达水平与胃癌进展呈正相关,血清HGF可以作为反映胃癌生物学行为和判断预后的有效指标。Objective：We measured serum concentrations of hepatocyte growth factor （HGF） in 80 patients with gastric carcinoma and compared them with various clinicopathological characteristics, for evaluation of the significance of serum HGF lev els. Methods： Preoperative serum HGF levels were measured by an enzyme linked immunosorbent assay （ELISA） system in 80 patients with gastric cancer. Results： The mean value of serum HGF in 80 patients was significantly higher than that in healthy control subjects （2.94 ± 2.67 ng/ml vs. 1.02 ± 0.31 ng/ml, P = 0. 002）. An increased serum HGF levels were associated with stage progression. Serum HGF levels in stage Ⅰ patients were significantly lower than those in Ⅲ stage and stage Ⅳ patients （2.19 ± 2. 12 ng/ml,3.92 ± 3.45 ng/ml and 4. 13 ± 3.71 ng/ml, respectively）, and statistically significant differences were found between the serum HGF levels of stage Ⅰ and those of stage Ⅲ （P〈0.05） and stage Ⅳ patients （P〈0.05）. Pathological factor analyses demonstrated that the patients with lymph node metastases and liver metastases had significantly higher ser um concentrations of HGF （P = 0. 016 and P = 0. 009, respectively）. Conclusion：The mean serum level of HGF in gastric cancer patients is significantly higher than that in healthy control subjects. Over-expression of the serum HGF level has been correlated with higher tumor stages. These findings suggest that the measurement of serum HGF levels might be a useful indicator for monitoring tumor progression and prognosis in patients with gastric carcinoma.
