摘  要:目的:分离和纯化非肥胖糖尿病(NOD)鼠的造血干细胞。方法:运用针对小鼠干细胞表面特异表达的干细胞抗原-1(Sca-1),采用磁性细胞分选方法(MACS)分离NOD鼠骨髓造血干细胞,并应用流式细胞术(FACS)监测分选效果。结果:纯化后Sca-1+细胞纯度平均为83.01%,细胞回收率为52.48%~57.64%,细胞活力在90%以上。结论:MACS可以从NOD鼠骨髓中分离出Sca-1阳性造血干细胞,细胞纯度可达80%以上。Objective:To isolate and purify hematopoietic stern cell from non obese diabetic mice. Method:The bone marrow hematopoietic stem cells were isolated from non-obese diabetic mice by means of magnetic activated cell sorting(MACS) using the stem cell antige-1(Sca-1) expressed on the surface of hematopoietic stem cells. Flow eytometry(FACS) was used to monitor the efficiency of the isolation. Result:The purity of Sea-1^+ ceils amounted to 83.01% in average after the purification, and the recovery rate was from 52.48% to 57.64%, the cell viability was above 90%. Conclusion:MACS can be used to isolate the Sea-1^+ hematopoietic stem cells from NOD mice and the purity of the isolated cell proportion was more than 80%.
