摘  要:目的:观察马钱子散对骨折愈合过程中骨痂局部BMP含量表达的影响,分析其促进骨折愈合的机理。方法:45只新西兰兔随机分为随机分为A组(正常组)5只和B组(空白对照组)20只和C组(用药组)20只,其中B、C组为人工手术造成大白兔双桡骨10mm缺损的标准骨折模型。C组以马钱子散26.27mg/kg调于20ml蒸馏水分早晚2次灌胃;B组则用20ml蒸馏水分早晚2次灌胃。分别于骨折后第1、2、3、4及6周,取标本进行抗骨形态发生蛋白(BMP)单克隆抗体免疫组化染色,通过图像分析系统对BMP含量进行定量分析。并以B组和C组作为对照组来比较分析各组促进骨折愈合的能力。结果:C组骨折局部BMP含量明显高于B组(P<0.01),C组BMP高峰出现在第2周,B组出现在第3周。结论:在安全有效剂量范围内使用马钱子散内服,能加快骨折修复细胞的分化、增殖,诱导骨折局部BMP合成,使BMP峰值提前,加速兔创伤性骨折的愈合,提高骨折愈合质量。Objective:To investigate effect of nuxvomica powder on the local content of BMP expression in the process of fraction healing, and to explore the mechanism of nuxvomica powder in improving fracture healing. Methods: Forty five New Zealand rabbits were randomly divided into group A (normal group, 5 rabbits), group B (control group, 20 rabbits) and group C (treatment group, 20 rabbits). Standard fracture model of 10 mm defect in bilateral radials were set up via op- eration. Rabbits in group C received intragastric administration of nuxcomica powder (26.27 mg/Kg) dissolved in 20 ml distilled water twice a day, while rabbits in group B were treated with 20 ml distilled water twice a day. The samples were collected at 1, 2, 3, 4 and 6 weeks, and immunohistochemistry and image analysis system were used to measure the BMP content. Group B and C were regarded as control to analyze the effect of each group on fracture healing. Results:The BMP content in fracture location of group C were significantly higher than that of group B (P^0.05). The peak value of BMP in group C and B appeared at 2nd and 3rd week respectively. Conclusion:In the safe and effective dosage, nuxvomica powder can promote the proliferation of fracture repairing cells in callus, enhance the activity of osteoblasts and induce the BMP synthesis which accelerate the fraction healing.
