摘  要:目的对文献报道的嗅鞘细胞(OEC)纯化方法进行比较,寻找一种较优化的纯化方法,为细胞移植修复脊髓损伤实验研究提供高纯度的种子细胞。方法实验分别比较了3种纯化方法:差速贴壁法、差速贴壁法联合阿糖胞苷化学纯化法、差速贴壁法联合NGFRp75抗体纯化法纯化原代培养的新生SD大鼠的OEC,并对纯化的OEC的形态学和免疫学特性等进行观察。结果与另两种纯化方法相比,差速贴壁法联合NGFRp75抗体纯化法得到的OEC不仅纯度高而且细胞活性好。结论该实验找到了一种纯化OEC的较优化方法——差速贴壁法联合NGFRp75抗体纯化法,此法得到的细胞较符合细胞移植的要求,为下一步细胞移植修复脊髓损伤实验研究打下了细胞学基础。Objective To compare the various purification methods in the culture of newborn rat olfactory ensheathing cells (OEC) for finding the more optimial purification method and providing higher purity seed cells in cell transplantation to repair spinal cord injuring. Methods Three kinds of purification methods ie differential adherent, differential adherent combined with Ara-C and differential adherent combined with NGFRp75 antibody purification method for purifying primary cultured newborn SD rat OEC were performed. The morphological pattern and immune characteristics of purified OEC with differential purification methods were observed and compared in the study. Results Not only high purity but also good activity cells were obtained by the differential adherent combined with NGFRp75 antibody purification method comparing with the other two purification methods.Conclusion The optimized purification approach for OEC ie differential adherent combined with NGFRp75 antibody purification method, is found in this study. Purified by this method the yielded cells are more satisfactory for the requirement of cell transplantation. It establishes the cytological basis for the experimental study of cell transplantation in repairing the spinal cord injuring.
