摘  要:目的探讨芦荟大黄素抑制人口腔癌细胞生长和迁移的双重效应及其机制。方法对人口腔上皮癌KB细胞分别用2.5、5、10、20和40μmol/L的芦荟大黄素处理,对照组为含0.1%二甲基亚砜的培养液,分别用结晶紫实验和划痕损伤愈合迁移实验分析长效抗增殖作用和抗迁移效应,并用Western blotting方法检测蛋白激酶Ca和c—myc蛋白水平的变化。结果芦荟大黄素能长时间抑制KB细胞的生长;划痕损伤愈合迁移实验结果显示,药物处理3d后进入划痕区域的细胞数为(6.2±0.2)个/mm^2,而对照组为(33.5±0.4)个/mm^2,差异有统计学意义(P〈0.05);对照组细胞抑制蛋白激酶Cα和c—myc的水平较高,经芦荟大黄素处理后,均以剂量依赖方式下降。结论芦荟大黄素的抗增殖和抗迁移活性与抑制蛋白激酶C信号传导途径有关。Objective To investigate the anti-proliferation and anti-migration dual effects of aloeemodin on KB cells and its mechanisms. Methods KB cells were treated with 2. 5, 5, 10, 20 and 40μmol/L aloe-emodin. Crystal violet assay was used to determine the long-term growth inhibition of aloeemodin on human oral cancer KB cells. Scratch wound-healing motility assay was used to measure the antimigration effect. The protein kinase Cα and e-mye expression changes in protein levels were detected by Western blotting. Results A durable cell growth inhibitory effect of aloe-emodin on KB cells was found. Treatment of aloe-emodin resulted in the inhibition of cell migration. The protein kinase Cα and c-mye in protein levels were decreased upon treatment with aloe-emodin compared with controls. Conclusions The anti-proliferation and anti-migration effects of aloe-emodin on KB cells are associated with the suppression of protein kinase C pathway.
