摘  要:以白桦(Betula platyphylla Suk.)组培苗茎段为研究材料,在IS培养基上附加0.6mg/LNAA+0.8mg/L BA进行愈伤组织的诱导,观察不同光处理条件下白桦愈伤组织的生长状况及其三萜积累的变化。结果表明:在一个继代生长期中,白桦愈伤组织在不同光照条件下的干质量积累呈上升趋势,其干质量增长率依次为:蓝光(48.95%)〉黄光(40.24%)〉绿光(23.28%)〉黑暗(21.14%)〉红光(18.37%);全光照(36.56%)〉14h光照(24.69%)〉黑暗(21.14%)。白桦愈伤组织的细胞活力在一个继代培养周期中呈现先上升后下降趋势,其中,愈伤组织的细胞活力在9-15d间达到最高值;白桦愈伤组织中三萜质量分数在0-9d呈上升趋势,9-18d呈下降趋势,18-21d略微回升,于第9d达到最大值。在第9d时,光处理下白桦愈伤组织中的三萜质量分数与对照(黑暗)相比的增长率依次为:蓝光(20.12%)〉红光(17.03%)〉黄光(9.35%)〉绿光(-3.09%);14h光照(12.06%)〉全光照(-3.01%)。由此可见,光处理可以调节白桦愈伤组织的生长状态及其三萜等次生代谢物的积累量。The stems of tissue-cultured seeding of birch (Betula platyphylla Suk. ) were cultured on IS media supplemented with 0.8 mg/L BA and 0.6 mg/L NAA to induce callus. The induced calluses were used to detect the rate of increased dry weight, cell viability and triterpenes content under different light conditions. During one subculture period, the rate of increased dry weight in callus under different light conditions exhibited blue light (48.95%) 〉 yellow light (40.24%) 〉 green light(23.28% ) 〉 dark(21.14% ) 〉 red light(18.37% ) and full light(36.56% ) 〉 14 h/d (24.69%) 〉 dark (21.14%). Under different light conditions, cell viability of callus was variable during one subculture period, and the highest cell viability occurred during the 9th day and the 15th day. The content of triterpenes showed an increasing tendency during the first nine days, a decreasing tendency during the 9th day and 18th day, and a slightly ascent during the 18th day and the 21st day. Triterpenes content reached the maximum value on the ninth day. Compared with the control (dark), the rate of increased triterpenes content displayed blue light(20. 12% ) 〉 red light(17.03% ) 〉 strong light (16.05%) 〉 14h/d(12.06% ) 〉 yellow light(9.35% ) 〉 full light( -3.01% ) 〉 green light( -3.09% ). Thus light treatment could regulate the growth of callus and the accumulation of triterpenes in birch.
