摘  要:目的:观察复方蒲黄颗粒(PH)对局灶性脑缺血再灌注大鼠神经元凋亡的影响,并探讨其作用机制。方法:采用大脑中动脉线栓法建立局灶性脑缺血再灌注大鼠模型。75只大鼠分为假手术、模型和PH高、中、低剂量组。采用Nissl染色观察神经元形态变化,TUNEL法检测神经元凋亡,原位杂交检测Caspase-3mRNA的表达。结果:PH高、中剂量组少量细胞核固缩深染,细胞丢失、凋亡细胞数及Caspase-3mRNA阳性细胞数均显著减少。结论:PH可下调Caspase-3的表达,从而抑制神经元凋亡,对脑缺血再灌注损伤有保护作用。OBJECTIVE: To investigate the protective effects of Compound Puhuang granules(PH) on neural apoptosis in rats with focal cerebral ischemia reperfusion, and to discuss the possible mechanism of its neuron protection.METHODS: The rats were surgically prepared for occlusion of the middle cerebral artery' (MCAO) according to the technique of Zea - Longa et al .Totally 75 SD rats were randomized into 5 groups(n = 15): sham operation(Sham) group; ischemia/ reperfusion(I/ R) group;PH of high dose (PHH) group; PH of medium does(PHM) group;PH of low dose(PHL) group. The morphologic changes of neurons were observed by using Nissl staining; the neuron apoptosis was detected by TUNEL method and the expression of Caspase - 3 mRNA were examined by using in situ hybridization.RESULTS: In PHH and PHM groups, karyopyknosis anachromasis was noted in small percentage of cells, and the cell loss, the number of apoptotic cells and the number of Caspase- 3 mRNA positive cells in the two groups were all significantly reduced.CONCLUSION:PH could inhibit neural apoptosis in rats with focal cerebral ischemia - reperfusion through down - regulating the expression of Caspase - 3 mRNA so as to achieve a protective effect on cerebral ischemia reperfusion injury.
