摘  要:通过体细胞核移植技术获得重构胚胎,在卵母细胞内多种重编程因子的作用下,供体细胞核抹去分化状态。恢复全能性,即核重新编程的过程。在这个过程中,供体核必须被正确激活与胚胎发育密切相关的基因,同时抑制与分化有关的基因。到目前为止,通过该项技术已经获得了多种哺乳动物的克隆后代,但克隆成功率低,且存活个体大多表型异常。核重编程的不准确性可能是克隆后代存在高死亡率和发育异常的主要原因之一。基因组DNA甲基化、组蛋白乙酰化状态是影响重编程的重要因素。该文主要就组蛋白乙酰化的生物学功能,组蛋白去乙酰化酶抑制剂TrichostatinA对组蛋白乙酰化状态的影响.以及TrichostatinA在体细胞核移植中的应用作以综述。Nuclear reprogramming is a course that differentiation state of donor nucleus is excelled and its totipotentiality is restored under actions of many reprogramming factors in the oocyte by using somatic cell nuclear transfer technique. In this course, the genes in the donor nucleus associated with embryonic development should be activated correctly, meanwhile, the genes associated with cell differenciation should be suppressed. Up to date, cloned offsprings of many kinds of mamalian animals have been achieved by using this technique. However, the success rate of cloning is low and majority of survival individuals show abnormal phenotypes. Inacuracy of nucleus reprogramming of somatic cell in enucleated oocyte may be one of important reasons why high death rate and developmental anomaly among coloned offsprings occur. Level of DNA methylation and histone acetylation in genome are important factors influcing nucleus reprogramming. In this paper, we reviewed biological functions of histone acetylation, influences of histone-deacetylase inhibitor trichostatin A (TSA) on histone acetylation and application of TSA in somatic nuclear transfer.
