摘  要:目的:探讨1,6-二磷酸果糖(FDP)对油酸致大鼠急性呼吸窘迫综合征(ARDS)治疗作用的可能机制。方法:将30只健康的Wistar大鼠随机分为对照组、模型组和治疗组,由股静脉注入药物,对照组注入生理盐水2.1mL/kg,ARDS模型组注入油酸0.1mL/kg和生理盐水2.0mL/kg,FDP治疗组注入油酸0.1mL/kg和用生理盐水稀释的FDP400mg/kg(2.0mL/kg),3h后测定血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)浓度,肺组织Na+-K+-ATP酶(NKA)活性,计算左肺湿/干重(W/D)比值,行肺组织病理学观察。结果:模型组与对照组比较,左肺W/D、血清中TNF-α、IL-1β浓度明显升高(P<0.01),而肺组织中NKA活性明显降低(P<0.01),肺组织出现明显病理损害;治疗组与模型组比较W/D、TNF-α、IL-1β、NKA改变较小(P<0.05或P<0.01),其肺损伤轻于模型组。结论:FDP可通过抑制TNF-α、IL-1β升高,提高肺组织NKA活性,对实验性ARDS大鼠起一定的治疗作用。Objective: To study the therapeutic effects of fructose-1, 6-diphosphate (FDP) on acute respiratory distress syndrome (ARDS) in rats induced by oleic acid (OA). Methods: Thirty healthy Wistar rats were randomly divided into three groups, control group, ARDS group and FDP-treated group. In control group normal saline (NS 2.1 mL/kg) was injected via femoral vein in rats. In ARDS group rats were induced ARDS by OA (0.1 mL/kg) and NS (2.0 mL/kg) injected via femoral vein. In FDP-treated group, OA (0.1 mL/kg) and FDP (400 mg/kg, diluted in 2.0 mL/kg of NS) solutions were injected via femoral vein respectively. After three hours, the contents of tumor necrosis factor-alpha (TNF-α), interleukin 1 β(IL- 1β) in serum, the activity of Na+-K+-ATPase (NKA) in lung tissue and the index(wet weight/dry weight)of left lung tissue were measured, and pulmonary pathological changes were compared between the three groups. Results: Compared with control group, the lung index in ARDS group showed a significant increase, as well as the levels of TNF-α, IL-1β in serum (P 〈 0.01). The activity of NKA in lung tissue showed a significant decrease (P 〈 0.01) and more typical pathological changes in lung tissue treated by OA. The changes in W/D, TNF-α, IL-1β, NKA, and pathological changes in FDP-treated group were less than those in ARDS group(P 〈 0.05 or P 〈 0.01 ). Conclusion: The results indicated that FDP inhibited the increase of IL-1β and TNF-α in serum, also increased the activity of NKA in lung tissue. FDP showed therapeutic effects on OA-induced ARDS in Rats.
