鸡传染性支气管炎(IB)诊断方法的研究

摘　　要：本试验用鸡胚大量繁殖鸡传染性支气管炎病毒（IBV）,收集尿囊液,用1％胰蛋白酶处理制备IBV血凝抗原,建立了血凝—血凝抑制（HA—HI）检测IBV抗体的方法,其特异性强、操作简便。另外,本试验建立Dot—ELISA方法用于检测IBV,病料经处理后,点样于硝酸纤维素膜,加兔抗IBV抗体,经PPA—DAB—H_2O_2系统显色,其检测结果与双抗体夹心ELISA的检测结果一致。两种诊断方法的建立为临床上检测病原和监测抗体提供了便捷的手段。Two diagnostic methods for IBV were established: the haemagglutination - inhibition test (HI) and Dot-ELISA.HI test was used for the detection of antibody to IBV, in the test.The haemagglutination antigen was prepared with crude allantoic fluid treated with 1% Trypsin, which showed a high HA litre to chicken red blood cell (RBC).In order to detect IBV,Dot-ELISA was developed, the sample was loaded on the nitrocellulose membrane, rabbit serum to IBV as primary antibody was then added, PPA was used as second antibody. The results showed that the assays had high sensitivity and good specificity.
