摘 要:目的观察清热解毒药配伍桔梗对慢性阻塞性肺疾病(COPD)大鼠模型病理形态学的影响,探讨桔梗在COPD治疗中的作用。方法采用气管注脂多糖加熏香烟联合造模方法建立COPD大鼠模型,设立假手术组、模型组及5个中药组(桔梗组、桔甘组、银翘组、桔银翘组、桔甘银翘组)。假手术组、模型组灌胃生理盐水,各给药组灌胃相应中药,连续28天。在光镜、电镜下分别观察不同组大鼠病理形态学的改变,并对病理切片进行半定量分析,观察各组肺平均内衬间隔(MLI)、平均肺泡数(MAN)变化。结果病理结果显示COPD模型组与假手术组相比肺组织结构严重受损,炎症细胞增多。桔梗组、桔甘组、银翘组对COPD病理改变与模型组相比均无明显差异;桔银翘组、桔甘银翘组与COPD模型组相比病理改变具有明显改善。半定量分析显示桔银翘组MLI较模型组有显著差异(P<0.05);桔甘银翘组MAN较模型组有极显著性差异(P<0.01)。结论清热解毒药物配伍桔梗,在COPD的治疗过程中能够发挥增效(引经)作用,增加甘草具有协同增效(引经)作用。Objective To observe the influence of Chinese herbal drugs with the effects of cleating away heat and removing toxin combined with Radix Platycodi on the pathomorphological changes in rats with chronic obstructive pulmonary disease (COPD), and to investigate the action of Radix Platycodi in the treatment of COPD. Methods The rat model of COPD was established by using the method of cigarette smoking combined with intratracheal injection of LPS. All rats were divided into the sham-operation group, model group and 5 Chinese herbal drug (CHD) groups (group 1 with Radix Platycodi, group 2 with Radix Platycodi and Radix Glycyrrhizae, group 3 with Honeysuckle and Forsythia, group 4 with Radix Platycodi, Honeysuckle and Forsythia, and group 5 with Radix Platycodi, Radix Glycyrrhizae, Honeysuckle and Forsythia). The sham-operation group, model group were given intragastrically the normal saline solution for 28 days, and 5 CHD groups were given corresponding herbal drugs intragastrically for 28 days. The pathomorphological changes in rats were observed by microscope and electron microscope in different groups. The changes of pulmonary mean lining intercept (MLI) and mean alveolar numbers (MAN) were observed after the semi-quantitative analysis on pathological sections. Results The results of pathology showed that the model group had more serious injury of lung tissue and more inflammatory cells compared with the sham-operation group. The pathological changes of COPD in group 1, 2 and 3 had no significant difference compared with those in the model group, but were relieved significantly in group 4 and 5. The results of the semi-quantitative analysis showed that MLI in group 4 had significant difference compared with that in the model group (P 〈 0.05 ) , and MAN in group 5 had great significant difference compared with that in the model group ( P 〈 0.01 ).
