摘  要:目的考察大孔吸附树脂分离纯化降香总黄酮的工艺条件及参数。方法选择9种不同极性的大孔吸附树脂,以降香总黄酮为指标,通过静态吸附和解吸附试验优选树脂,动态试验考察其纯化降香总黄酮的最佳工艺条件。结果静态试验选出HPD-100树脂,其最佳静态吸附时间和解吸附时间均为5 h,样品溶液浓度为3.7 mg/mL,吸附流速为2 mL/m in,10倍床体积的70%以上浓度乙醇洗脱,可解吸附70%的降香总黄酮。结论HPD-100树脂能够有效纯化降香总黄酮。Objective To study the purification process of total flavonoids in Lignum Dalbergiae odoriferae with macroporous resin.Methods 9 kinds of macroporous resins with different polarities were used in the purification of the total flavonoids in Lignum Dalbergia odorifera.Results HPD-100 macroporous resin was better in purification of the total flavonoids.The optimal static adsorbing time and desorbing time were both 5 h,the optimal concentration of sample was 3.7 mg/mL,flow speed was 2 mL/min,and eluted with ethanol at the concentration of more than 70% which volume was ten times of elution bed volume,what was better in eluotropic effect with smaller in tailing level.Conclusion HPD-100 macroporous resin could be used in the purification of the total flavonoids in Lignum Dalbergiae odoriferae.The adsorption quantity of total flavonoids at saturation was 24.983 mg/g.
