摘  要:新近研究发现,神经生长因子(NGF)对非神经系统也具有重要的功能效应。为探讨NGF与骨髓造血组织的关系,本实验制备地高辛标记的人βNGFcDNA探针,通过原位杂交技术,检测正常人骨髓NGFmRNA的分布。实验显示,βNGFcDNA探针与其互补的mRNA杂交反应信号存在于巨核细胞系和组织嗜碱细胞,杂交反应产物呈紫蓝色颗粒,分布于阳性细胞胞浆中。实验结果提示,NGF基因在骨髓造血组织也有表达,可能参与造血功能的调节。本实验制备的地高辛标记βNGFcDNA探针灵敏度高达0.1pg/ml,且特异性强、分辨力高、重复性好,是研究NGF荃因表达和调控的有力工具。Nerve growth factor (NGF) is an important bio-active substance of the nervous system.The recent research indicates that NGF and its receptor are also widely distributed in the non-nervous system and play an improtant role.In order to explore the relationship human β NGF cDNA probes which were labelled with 11-dUTP-digoxigenin and detected the distribution of NGF mRNA in normal human bone marrow by hybridization in situ method.Exprimental results showed that strong hybridized signal was localized in the megalocaryocytes and tissue basophilic cells. The reactive products of hybridization were blue and purple-blue granule and distributed in cytoplasm. Erythrocyte series and granulocyte series were negative.The results demonstrate that NGF gene also expressed in the hematopoietic tissue of bone marrow and NGF might play important role of hematopoietic functions.The NGF cDNA probes prepared in our laboratory have higher sensitivity,stronger specificity and better resolving power,which serve as good tools to study the expression and regulation of NGF mRNA.
