摘  要:补体系统过度活化产生的多种炎症介质通过多种机制参与了如缺血再灌注损伤、超急性排斥反应等疾病的细胞和组织损伤过程。因此,研究和开发出能拮抗补体过度活化的高效、安全的补体抑制剂是临床上亟待解决的问题。近年来,补体抑制剂的研究取得了长足的进展,文中就补体C3和C5相关抑制剂、小分子补体抑制剂、C1酯酶抑制剂及攻膜复合体抑制剂的研究进展做一综述。Over-activation of complement system produces inflammatory mediators that lead to the inflammatory response in immune-mediated cell and tissue damage through many pathways in many kincls of diseases, such as ischemia-reperfusion injury and hyperacute transplantation rejection. Therefore, developing complement inhibitors with high potency and safety has been an urgent problem in the clinical practice. In recent years, the research on complement inhibitors has aehieved remarkable advancement. This article gives a general review on several kinds of promising complement inhibitors, including C3/C5-related complement inhibitors, small molecule complement inhibitors, C1 esterase inhibitor and membrane attack complex (MAC) inhibitor.
