摘 要:目的:研究不同基团的甾体底物在不同反应体系中的生物脱氢规律及细胞壁、细胞膜对生物转化的影响。方法:将研究底物置于4种不同的反应体系中,即采用普通直接转化、双液相转化、破碎细胞转化和原生质体转化方法,测定各甾体的熔点、比旋度、吸光系数等理化性质,并通过非线性拟合方法建立了甾体化合物C1,2位脱氢转化率与其理化性质之间变化规律的数学模型。结果和结论:保证细胞的完整性对C1,2位脱氢转化至关重要;甾体底物在普通直接转化中,脱氢转化率按由高到底排列依次是醋酸可的松、普氏脱溴物、醋酸氢化可的松、甲睾酮、睾丸素、4-雄烯二酮和可的松,RSA不转化;4种反应体系按其甾体化合物转化率高低的排序依次是普通直接转化、双液相转化、破碎细胞转化和原生质体转化(醋酸氢化可的松除外)。在同一种反应体系中,数学模型显示不同底物的熔点和吸光系数对其转化率有着正向促进作用,而比旋度对其转化率具有负向抑制作用。Objective:To investigate the dehydrogenation of steroids with different groups in the different reaction systems, and the effect of the cell wall and cell membrane on biotransformation. Methods: The steroidal compounds were added to 4 of reaction systems, i.e. the general, biphasic, protoplast and ultrasonic cell biotrans- formation. Using Matlab, the relationship of function between the bioconversion rate and the melting point, specific rotation, and absorption coefficient were established. Results and Conclusion: It is very important to ensure the cell's integrality in the process of bioconversion. In the general biotransformation, the order of bioconversion rates (from high to low) of 8 steroidal compounds was cortisone acetate, debrominative substance, hydrocortisone acetate, methyltestosterone, testosterone, 4-androstenedione, cortisone and RSA (that was not transformed). The order of 4 reaction systems from high to low was the general biotransformation, biphasic biotransformation, ultrasonic cell biotransformation and protoplast biotransformation (except hydrocortisone acetate). In the same reaction system, the mathematic model showed that the melting point and absorption coefficient promotes the bioconversion rate, and the specific rotation restrains the bioconversion rate.
