摘  要:[目的]筛选与纯化棉花的优势内生菌。[方法]以棉花幼苗为试材,表面彻底消毒后,取组织匀浆涂布于不同培养基平板,观察培养出的菌落形态、挑取不同形态的菌落,然后进行平板划线纯化、镜检、斜面低温保存和菌种鉴定。[结果]不同灭菌时间对棉花幼苗具有不同的灭菌效果。灭菌时间越长,灭菌效果越好,杂菌越少。灭菌8 min较合适。分离后在固体琼脂培养基上长出了不同特征的单菌落,其菌落形态和颜色有所差异。有表面光滑的,有表面萎缩的,有周边圆润的,有周边呈不规则形状的。对所标记的菌落进行革兰氏染色,部分菌落被染成红色。[结论]芽孢杆菌为棉花内生菌的优势种群,它的分离频率最高。[ Objective] The aim was to select and purify the dominant endophytic fungus from cotton. [ Method] With cotton seedlings as the tested material, after their leave surfaces were thoroughly disinfected, their tissue homogenate was coated on different medium plate, the morphologies of the cultured colonies were observed and the colonies with different morphologies were selected. After that, the colonies were purified with streak plate, examined by microscopic and preserved on inclined plane with low temperature and their strains were identified. [ Result ] The sterilization with different times had different sterilization effects on cotton seedlings. The longer the sterilization time was, the better the sterilization effect was and the less the mixed bacterium was. The sterilization time of 8 rain was better. After separation, the single colonies with different characters grew out in the solid agar medium and the colonial morphologies and colors were different. There were the colonies with smooth surface, atrophy surface, round peripheral and irregular shape of peripheral. After the labeled colonies were dyed with gram staining method, they were partly dyed red. [ Conclusion] The bacillus was the dominant endophytic fungus in Xinjiang cotton and its isolation frequency was the highest.
