摘 要:以韭莲组培苗鳞茎为试验材料,通过正交试验筛选出最佳的小鳞茎增殖培养基:MS+6-BA 0.5 mg/L+NAA 1.5 mg/L+蔗糖50 g/L.将诱导得到的小鳞茎苗转入添加了不同激素和蔗糖浓度的1/2MS培养基上,得到最适生根培养基为1/2MS+NAA 0.5 mg/L+蔗糖15 g/L,移栽成活率100%.利用RP-HPLC法测定组培苗和野生苗鳞茎中石蒜碱和加兰他敏的含量,韭莲组培苗鳞茎中石蒜碱和加兰他敏的含量分别为204.83μg/g,98.7μg/g;野生苗鳞茎中石蒜碱和加兰他敏的含量分别为225.36μg/g,63.72μg/g.组培苗鳞茎中加兰他敏的含量是野生苗鳞茎的1.55倍.The bulbs of Zephyranthes grandiflora were used as explants in tissue culture. According to the results of orthogonal test L9 (3^4 ), the optimal medium for bulb multiplication was identified to be MS +6-BA 0. 5 mg/L+NAA 1.5 mg/L+sucrose 50 g/L. The optimum medium for rhizogenesis was 1/2MS +NAA 0.5 mg/L+sucrose 15 g/L, and a survival rate of 100% was achieved after the transplantation of the rooted plantlets. Determination by RP-HPLC showed that the contents of lycorine and galanthamine were 204.83 μg/g and 98.7μg/g for the tissue culture-derived plantlets, respectively, or the content of galanthamine was 1.5g times for wild Z. grandiflora (225.36 μg/g and 63.72 μg/g).
