摘 要:目的探讨实验性糖尿病(DM)大鼠肺组织晚期糖基化终末产物(AGEs)、转化生长因子-β1(TGF-β1)、结缔组织生长因子(CTGF)的变化及其相关性。方法48只SD雄性大鼠随机分为DM组和对照组,每组24只。链脲佐菌素(STZ)60mg/kg尾静脉注射制造DM模型。分别于造模成功后4、12、20周末取材。免疫组织化学方法观察肺组织AGEs、CTGF、TGF-β1的变化,并进行图像分析。结果DM大鼠肺泡上皮细胞、支气管黏膜上皮细胞、血管内皮细胞及平滑肌细胞可见大量AGEs阳性表达,平均灰度值低于对照组(P<0.05)。随着DM病程的发展,AGEs阳性表达逐渐降低(P<0.01);4周DM大鼠支气管黏膜上皮细胞、血管内皮细胞、肺间质细胞可见少量CTGF、TGF-β1阳性细胞,12、20周时可见大量CTGF、TGF-β1阳性细胞,平均灰度值均低于对照组(P<0.01);Pearson相关分析显示,DM大鼠肺组织AGEs与CTGF、TGF-β1之间呈显著正相关(γ=0.939、0.904;P<0.01),CTGF与TGF-β1亦呈正相关(γ=0.878;P<0.01)。结论DM大鼠肺组织内AGEs随病程的延长其表达明显增加,并与CTGF、TGF-β1正相关。Objective To approach the changes of advanced glycosylated endproducts (AGEs), transforming growth factor(TGF-β1) and connective tissue growth factors(CTGF) in the lungs of experimental diabetic rats and their relationship. Methods Totally 48 male SD rats were divided into the diabetes mellitus(DM) group and the control group,with 24 rats in each group.The DM rat model was made by the injection of streptozocin (60mg/kg) into the caudal vein. The rats were killed and the lungs were taken at the end of the 4 th, 12 th and 20th weeks respectively after the models were established. The changes of AGEs, CTGF, TGF-β1 in rat lungs were observed with immunohistochemical assay and the image was analyzed. Results A great quantity of AGEs positive cells were observed in the alveolar epithelial cells, bronchial mucosal epithelium, anglo-endothelial ceils and smooth muscle cells of the DM rats. The average gray(AG) was lower than that of the controls( P 〈 0.05) and decreased with the DM course( P 〈 0.01) . In the 4-week DM rats, there were a few CTGF and TGF-β1 positive cells in the bronchial mucosal epithelium, angio-endotheliial ceils and lung interstitial ceils. In the 12-and 20-week DM rats, there were a great many CTGF and TGF-β1 positive cells. The AG was lower than that of the controls( P 〈 0.01 ). Pearson correlation analysis revealed that AGEs were positively related with CTGF, TGF-β1( γ= 0.939,0.904; P 〈 0.01 ), and so was the case with CTGF and TGF-β1 ( γ = 0. 878 ; P 〈 0.01 ). Conclusion The AGEs in the DM rats lung increased obviously with the DM course and positively correlated with CTGF, TGF-β1
