摘 要:以富士、乔纳金、金矮生、嘎拉组培苗为试材,在继代培养基中加入不同浓度的比久、矮壮素、多效唑及脱落酸,研究了植物生长延缓剂和生长抑制剂对苹果组培苗种质延缓生长保存的效应。结果表明:比久、矮壮素均有明显抑制苹果组培苗生长的作用,且茎尖存活率高,试管苗生长状况较好,转入常规继代培养基当代即可恢复生长,可用于苹果种质资源的离体保存,比久适宜的浓度为80mg/L,矮壮素为75mg/L左右;多效唑处理虽能明显抑制苹果试管苗生长,但易使有些品种试管苗玻璃化,可选择性应用;脱落酸作用过于剧烈,绝大部分处理苗死亡,不宜在苹果种质保存中使用。Daminozide (Bg), chlormequat chloride (CCC), pacl supplied in culture media during subculture process in apple in order to study the efficacy of plant growth regulators on obutrazol (PP333), and abscisic acid (ABA) were cultivars of Fuji, Jonagold, Goldspur, and Gala slow growth of apples in vitro for germplasm conservation. The results showed that B9 and CCC could significantly retard the growth of plantlets with a high surviving rate and healthy growth status, and the plantlets could resume their transferred onto routine subculture medium. The optimal concentration for B9 was growth immediately after 80mg/L and for CCC was about 75 mg/L, which could be used in apple germplasm conservation in vitro. CCC could also significantly retard the growth of plantlets, but that could cause vitrification in some cuhivars, so its application was selective with caution. ABA had a very strong effect and could result in death for most of the plantlets, and then its application was not suggested in apple germplasm conservation in vitro.
