摘  要:目的体外分离犬骨髓间充质干细胞,经诱导分化形成内皮细胞,构建生物人工血滤器,观察内皮细胞生长特点。方法骨髓间充质干细胞采用体外离心法分离培养,经诱导分化后形成内皮细胞,经传代殖后在中空纤维滤器内生长,建立生物人工血滤器,并对急性肾衰竭犬进行体外滤过实验。结果中空纤维滤器内灌注细胞浓度1.2×107/ml,内皮细胞的活性为95%,经培养10d后,平铺在中空纤维滤器内表面。经体外透析4h后,治疗组、对照组血清BUN(μmol/L)分别为88.4±6.1,141.5±6.2,差异具有统计学意义(P<0.01),钾、钙的差别无统计学意义。治疗组、对照组滤过液/血清BUN(μmol/L)分别为0.91±0.08,0.96±0.04,差异无统计学意义(P>0.05)。总蛋白与白蛋白的差异有统计学意义(P<0.01),实验组平均滤过液/血清总蛋白、平均滤过液/血清白蛋白分别是0.040±0.008、0.0034±0.0007,有选择性滤过功能。结论初步证实生物人工血滤器可以体外构建,并有一定的选择滤过功能。Objective To in vitro separate canine hone marrow-derived mesenchymal stem cells (MSCs) and to be induced as endothelial cells which form bioartificial hemofilter. Then to observe the growth characteristics of endothelial cells. Methods MSCs were separated by density gradient centrifugation and induced to form endothelial cells, and then cultured in the inner lumina of the fibre hemofiher,so the bioartificial hemofilter was constructed and the further experiments were performed on dogs with renal failure. Results The infused cells density in the hollow fibers was 1.2 × 10^7/ml, and the cell activity was 95 %. After being cultured for ten days,the cells in the fiber were in hautehen state. After 4 hours of dialysis, sera BUN (μmol/L) in experimental and control groups were 88.4± 6.1,141.5±6.2 respectively,wihich were significantly different (P〈0. 01),while there was no significant difference on the contents of K^+ and Ca^2+. The fluid-toserum BUN ratios in experimental and control groups were 0. 91±0.08 and 0.96±0. 04. There was no statistical difference (P〉0.05). In contrast to BUN, the significant differences were seen in all groups for total protein and albumin (P〈 0. 01). The mean fluid/serum total protein ratio was 0. 040±0. 008,while the mean fluid/serum albumin ratio was 0. 003 4 ±0. 000 7 for the experimental group. Conclusion These results provide the initial proof of concept in developing a tissueengineered hemofiher prototype, which has size-selective filtrate function.
