摘 要:目的分析海南省2005年霍乱菌株暴发分离株的分子特征及菌株之间的分型相似性,为防治工作提供病原依据,并与PulseNet China提供资料进行相关分析。方法对分离株进行噬菌体-生物分型,通过PCR检测霍乱毒素基因,利用脉冲场凝胶电泳(PFGE)进行分子分型和相似性聚类分析。结果54株分离株中有51株为产毒株,稻叶血清型,噬菌体-生物分型为1b型,其余3株为非流行株。全部菌株被分为15种PFGE带型,聚类分析大部分产毒株之间高度相似,并与PulseNet China提供资料证实,与同时福建、广东、浙江流行的稻叶型菌株带型相同。结论海南省2005年稻叶型暴发菌株,属于2005年流行的菌型,提示在我国东南沿海地区的广泛扩散。Objective To investigate the molecular characteristics and molecular typing of cholerae strains collected in Hainan Province in 2005 and analyze the typing cluster compared to the similar isolates in other provices at the same peroid. Methods Phage-biotyping was used to identify the biotypes of the isolates, toxigenicity was confirmed with ctxAB gene ampification Pulsed-field gel eleetrophoresis (PFGE) was performed in the molecular subtyping of isolates, the patterns were analysed with BioNumerics software to perform cluster analysis. The data was presented to PulseNet China database to search the matched isolates from other provinces. Results Within the 54 isolates, 51 of Inaba serotype were toxigenic, and Phage-biotype was lb. The other 3 isolates were nontoxigenic and belonged to nonepidemic strain based on Phage-biotyping scheme. Fifteen distinctive PFGE patterns were identified in all the 54 isolates. PFGE patterns of most toxigenic strains were identical or highly similar, and matched with the epidemic isolates in Fujian, Guangdong and Zhejiang in 2005. Conclusion The isolates from an outbreak of cholera in Hainan are highly homology, and belong to the cluster of 2005 epidemic strains from other provinces, suggesting the spread of the strains with such pattern in Southeast China in 2005.
