摘 要:目的研究尘螨主要变应原组分及性质。方法用SDS-PAGE、Western印迹和LC-MS/MS方法,对屋尘螨和粉尘螨的主要变应原组分进行筛选、分析和鉴定。结果两种尘螨的三个样本均有数条在蛋白大小和含量方面较为独特的条带,其中屋尘螨和粉尘螨抗原间蛋白质构成差别较大,两个粉尘螨样本间蛋白带差别较小。屋尘螨主要变应原大小约为12~15kDa和94~98kDa;粉尘螨主要变应原大小约为49~53kDa和94~98kDa,但其中一粉尘螨样本在49~53kDa和94~98kDa位置却未出现反应带。用LC-MS/MS方法从主要变应原条带中鉴定出屋尘螨变应原蛋白约64个,粉尘螨变应原蛋白约84个。它们包括一些已知的尘螨变应原、酶类、translation elongation factor、ribosomal protein、外膜(脂)蛋白、Keratin、Glycine cleavage system protein、binding protein、Heat shock protein、regulation protein、hypothetical protein、pol polyprotein和pol protein等蛋白质类别。结论屋尘螨和粉尘螨主要变应原构成及其免疫学反应强度均存在一定区别。针对中国人的尘螨主要变应原可能包括Der p2、Der p14、Der f3,Der f15和Der f17等,上述鉴定的其它蛋白质的一些也可能是潜在的变应原。Objective To screen and identify the dominant allergens relative to Chinese in house-dust mites Methods With SDS-PAGE, Western blotting and LC-MS/MS methods, the dominant allergens have been identified and analyzed from extracts of Dermatophagoides pteronyssinus (D.p) and Dermatophagoides farina (D.f). Results There were some unique protein bands in quantity and molecular size among three samples of D.p and Df . Composition of proteins had more differences between D.p and Df samples, less differences between two Df samples. Molecular size of the dominant allergens (protein) of D.p were about 12-15kDa and 94-98kDa, Df about 49-53kDa and 94-98kDa. In Western blotting, a Df sample had 49-53kDa and 94-98kDa allergens reaction bands, but in another sample they could not be seen. With LC-MS/MS methods, 64 allergen proteins of D.p and 84 allergen proteins of Df were identified from dominant allergen bands, including some house-dust mite allergens reported, many kinds of enzymes,translation elongation factor,ribosomal protein,outer membrane (lipo-)protein,keratin,glyeine cleavage system protein,binding protein,heat shock protein,regulation protein, hypothetical protein,pol polyprotein and pol protein etc. Conclusion There are differences in composition and immunoreactivity of dominant allergens of D.p and Df. The dominant allergens relative to Chinese in D.p and D.f may include Der p 2 (group 2 allergen of Dermatophagoides pteronyssinus), Der p 14, Der f 3, Der f 15 and Der f 17 et al. Some proteins mentioned above from D.p and Dfmay be potential allergens.
