摘　　要：目的研究自身免疫性睾丸炎睾丸病理形态和TNF-α在睾丸中的表达情况,探讨TNF-α在睾丸炎进展中的作用及机制。方法参照相关方法建立经典的自身免疫性睾丸炎模型,免疫80d后取材,HE染色观察睾丸组织结构变化,免疫组织化学方法定位TNF-α在睾丸组织中的表达,ELISA法测血清TNF-α含量。结果模型组生精上皮坏死、脱落,睾丸间质水肿,大量炎症细胞增生浸润。TNF-α在睾丸炎间质细胞胞浆、生精小管基底膜、多核巨大精细胞呈强阳性表达,生精上皮表达较弱,且模型组血清TNF-α含量显著高于正常对照组(P<0.05)。结论过量的TNF-α影响生精上皮的正常发育,参与自身免疫性睾丸炎的进展。Objective To observe the testicular pathologic morphology and expression and localization of TNF-α in EAO of rats and explore its effects on orchitic progression and its mechanism. Methods Duplicated EAO model, Sprague- Dawley rats were divided randomly into control group and model group. Eighty days ager immunization the levels of serum TNF-α were determined, the changes of testicular structure were observed by light microscope, localizations of expression of TNF-α were made by immunochemistry （IHC）. The contents of serum TNF-α was determined by ELISA. Results The spermatogenic epithelia of model group was degenerated and depleted,the testis interstitial edema,the proliferating and invading inflammatory cells were oberved under hght microscope. Expression of testicular TNF-α was positive significantly in the interstitial cells and proliferating inflammatory cells of testes, the level of plasma TNF-α in model group was significantly higher than normal group （P〈0.05）. Conclusion Excessive expression of TNF-α might affect normal development of seminiferous epithelium and involves in orchitic immune progression.
