摘 要:目的探讨一氧化氮参与卵泡发育和凋亡的机制。方法采用免疫组化的方法检测NO供体硝普钠(SNP)对成熟大鼠卵巢中凋亡调控蛋白Bcl-2、Bax表达的影响。结果实验(SNP)组Bcl-2蛋白在窦前卵泡、窦状卵泡中表达下调,Bax蛋白在始基卵泡、窦前卵泡、窦状卵泡中的表达明显上调,与对照组比较差异有显著性(P<0.05)。结论高浓度NO可上调大鼠卵泡中Bax蛋白的表达,下调Bcl-2的表达。推测NO可能通过Bcl-2/Bax途径来诱导卵泡中的细胞凋亡及卵泡闭锁。Objective To investigate the mechanism of nitric oxide (NO) participating in follicular development and apoptosis. Methods Immunohistochemistry was conducted to analyze the effect of NO donor (SNP) on the expression of apoptosis-related protein Bcl-2/Bax in the ovary of maturate rats. Results Compared to normal control group,the expression of Bcl-2 proteins is significant downregulation in preantral follicles and antral follicles of SNP group,the expression of Bax proteins was significantly upregulated in primordial follides and preantral follicles and antral follicles of SNP group. Conclusions The increase of NO concentration may up-regulate the expression of Bax protein and down-regulate the expression of Bcl-2 protein in follicles of rat ovary. Bcl-2/Bax system might be involved in NO to induce cell apoptosis in follicle and follicular atresia.
